The Irishman maintained his sparkling run of form with a treble at Ayr on Monday.
James Bethell's mare, who has won her last two starts and carries the colours of her Perthshire owner/breeder Robert Gibbons, faces just three rivals in the Kilmany Cup Handicap Chase.
Hughes said: "Thankyou Very Much is in great form after her wins at Leicester and course and distance.
"She is very well and there is no problem with the track, the trip or the ground.
"There is no reason why she shouldn't go close again as she has taken very well to fences."
It's almost exactly 30 years to the day since Musselburgh's jumps circuit was opened on January 5, 1987 and Chris Grant, who rode on the card that historic day, trains Hughes' fifth mount, Robins Legend.
The Teesside raider is equipped with a first-time visor as he makes his handicap debut over a longer trip.
Hughes' ride in the bumper, Kevin Ryan-trained newcomer Beyond The Clouds, has a Classic pedigree as he is a son of 1997 Arc hero Peintre Celebre.
